The purpose of the current study was to investigate pharmacodynamic interactions between vancomycin, amikacin, ofloxacin and trimethoprim/sulfamethoxazole (TMP-SMX) for the treatment of human infections. Serum and dialysate concentrations were measured in in vitro dialysis studies. The data for the in vivo dialysis studies were subjected to a noncompartmental pharmacokinetic analysis. Clinical data were used to evaluate the data from the in vitro dialysis study. A computer program was used to identify potential pharmacodynamic interactions between the four drugs and/or the four drugs and the dialysate. Potential pharmacodynamic interactions were evaluated according to the mechanism of action of the antibiotic, regardless of the elimination pathway. The predicted potential effects included either antagonism or synergism for the in vitro dialysis studies. In the in vivo dialysis studies, the predicted effects included synergism for vancomycin, amikacin, and ofloxacin, and antagonism for TMP-SMX. In the clinical study, the predicted interactions included an increase in the area under the concentration versus time curve for ofloxacin and vancomycin, and antagonism for TMP-SMX and amikacin. The clinical implications of the pharmacodynamic interactions were that vancomycin and ofloxacin may require dosage adjustment during concurrent use with TMP-SMX. These results should be interpreted cautiously, however, because amikacin and TMP-SMX were studied clinically under different conditions and the predicted interactions were based on different criteria. Further study is needed to confirm the clinical significance of these pharmacodynamic interactions.Gender-specificity in the pharmacokinetics of the first-in-class, novel, anti-malarial mefloquine in healthy Chinese subjects. The objective of the study was to characterize the pharmacokinetics of the novel, anti-mal be359ba680
